NHS-esters react with primary amines (N-termini of proteins) under physiological conditions to form a covalent amide bond releasing N-hydroxysuccinimide. Typical NHS-ester reactions are performed between pH 7-9 at 4 deg C to room temperature from 30 minutes to 2 hours.
